摘要 目的:建立以高效液相色谱法测定不同产地鬼臼类药材中鬼臼毒素含量的方法。方法:色谱柱为Diamonsil C18(250mm×4.6mm,5μm),流动相为甲醇-0.1mo·lL-1磷酸二氢钾(45∶55),检测波长为290nm,柱温为30℃,流速为1.0mL·min-1。结果:不同产地鬼臼类药材所含鬼臼毒素的量差异较大,其中以产自湖北竹溪的山荷叶含量最高,产自湖北房县的六角莲含量最低(二者含量相差近4倍)。结论:不同产地、不同品种的鬼臼类药材中鬼臼毒素含量不同,故各地的中毒剂量不同。 OBJECTIVE:To establish HPLC method for the content determination of podophyllotoxin in podophylloideae collected from different habitats.METHODS:Diamonsil C18(250 mm×4.6 mm,5 μm) was used with methanol-0.1 mol·L^-1 KH2PO4(45:55) as mobile phase at a flow rate of 1.0 mL·min^-1.The detection wavelength was set at 290 nm and column temperature was 30℃.RESULTS:The contents of podophyllotoxin in podophylloideae form different habitats were different from one another,among which the content of podophyllotoxin in Diphylleia sinensis from Zhuxi in Hubei province was the highest while that in Dysosma pleiantha from Fangxian in Hubei province was the lowest (difference between two kinds of plants of nearly 4 times).CONCLUSION:The contents of podophyllotoxin are different because of the different species and regions.The dose of podophyllotoxin which induce toxicosis is different.
