醒神解毒方预处理对全脑缺血再灌注大鼠海马神经元HSP70表达的影响 被引量:2

摘要 目的:探讨醒神解毒方预处理减轻全脑缺血再灌注大鼠海马神经元损伤的机制。方法:实验选用20只雄性SD大鼠,随机将20只大鼠分为假手术组、药物预处理组、缺血预处理组、脑缺血再灌注组,采用热凝椎动脉,钳夹双侧颈总动脉建立全脑缺血模型,假手术组暴露双侧颈总动脉不夹闭;缺血再灌注组,夹闭双侧大脑颈总动脉10 min后再灌注;缺血预处理组预缺血3 min,3 d后给予缺血10 min后再灌注24 h后处死;药物预处理组,灌胃2周后,作缺血再灌注处理,免疫组织化学染色观察HSP70的表达。结果:药物预处理及缺血预处理缺血区HSP70表达与与缺血再灌注比较,P<0.01。结论:药物预处理对随后的脑梗死有明显的保护作用,能诱导缺血耐受的产生。 Objective :To investigate the preconditioning effects of Xingshen Jiedu Decoction and the effect of ischemic preconditioning on the changes of structure. Method:Twenty male SD rats( weight 180 - 220 g)were randomly divided into 3 groups: sham operation group ( n = 5 ) ; the skin was sutured after common carotid artery( CCA ) was exposed ; ischemic group( n = 5 ) ; ischemic preconditioning group ( n = 5 ) Xingshen Jiedu Decoction pretreatment group ( n =- 5 ). Global cerebral ischemia and reperfusion model were established with Pulsinelli' s method. Result : Compared with ischemic group, the HSP70 expression of hippocampal CAl region was increased significantly in Xingshen Jiedu Decoction pretreatment group and ischemic preconditioning group( P 〈 0. 05 ). Conclusion : Ischemic preconditioning can obviously increase brain tissue tolerance of the brain to secondary severe ischemia.
