淫羊藿苷对SD大鼠成骨-破骨细胞共育体系的影响 被引量:16

摘要 目的:检测成骨-破骨细胞共育体系中应用淫羊藿苷后碱性磷酸酶(ALP)和抗酒石酸酸性磷酸酶(TRACP)的变化,探讨淫羊藿苷的作用机理。方法:建立成骨-破骨细胞共育体系,分为淫羊藿组、空白对照组和阳性对照三组,每组于同一培养板重复6孔,分别检测上清液中ALP和TRACP的含量,并进行组内和组间的统计学分析。结果:经鉴定成功培养成骨细胞和破骨细胞,并建立成骨-破骨细胞共育体系,测定结果显示,在ALP活性中,三组间比较差异显著(P<0.01),LSD法组内比较淫羊藿苷组与空白对照组差异显著(P<0.01);淫羊藿苷组与阳性对照组无显著性差异(P=0.546);在TRACP活性中:三组间比较差异显著(P<0.01),LSD法比较淫羊藿苷组与空白对照组比较差异显著(P<0.01);淫羊藿苷组与阳性对照组无显著性差异(P=0.10)。结论:淫羊藿苷可以增强共育体系中成骨细胞的活性,抑制破骨细胞的功能。 Objective:To check the level change of ALP and TRACP after-using icariin on osteoblast and osteoclast co-cultured system and study the mechanism of icariin. Methods:To establish osteoblast and osteoclast co-cultured system and then divided into three groups, icariin group, blank group and positive control group and each group repeated six times in same condition. The level of ALP and TRACP had been checked, the data. had been analyzed between and in groups by SPSS13.0. Results:After identification, the culturing of osteoblast and osteoclast were successful and the co-cultured system was established too. The checking result of ALP activity displayed that there was statistics difference among three groups (P 〈 0. 01 ). There was statistics difference between icariin group and blank group ( P 〈 0.01 ) and that was on difference between icariin group and positive control group ( P = 0. 546). The checking result of TRACP activity displayed that there was statistics difference among three groups (P 〈0.01 ) too. There was statistics difference between icariin group and blank group (P 〈0.01 ) and that was on difference between icariin group and positive control group (P = 0. 10). Conclusions :Icariin can enhance the activity of osteoblast and inhibit that of osteoclast in co-cuhured system.
