血清S-100ao蛋白测定及在心肌疾病中的应用

Determination of S-100ao protein in serum and its application in cardiac muscle disease

建立血清S-100ao蛋白测定方法并探讨其在心肌疾病中的应用。方法 以S-100ao蛋白纯品免疫家兔,得抗S-100ao蛋白抗体,用双抗体夹心酶联免疫吸附测定血清S-100ao蛋白。结果 方法线性可达10μg/L,批内变异为5.4％,批间变异为7.0％,平均回收率为98.6％。急性心肌梗死(AMI)早期S-100ao蛋白明显增高,高值持续2周以上。心绞痛及陈旧性心肌梗死S-100ao正常。结论 S-100ao测定可作为AMI的早期诊断、鉴别诊断及疗效评价的有用临床指标。

To establish a method to measure S-100ao protein and to study its application in cardiac muscle disease. Methods Rabbits were immnunized with S-lOOao protein.The method was a 'sandwich' ELISA for determining concentration of S-lOOao protein in serum. Results Within-run and between-run CVs were 5.4% and 7.0%,respectively. Analytical recovery averaged 98.6%. S-lOOao protein in serum was obviously higher in the easly phase of AMI and remained high in the late phase of AMI. S-lOOao protein remained in the normal range in the patients with angina pec-toris and old myocardial infarction. Conclusion serum S-100ao protein is a useful marker for the diagnosis in the early phare and differential diagnosis of AMI and evaluation curative effect of AMI.
