摘要
目的建立大鼠慢性胰腺炎模型,检测模型大鼠胰腺组织转化生长因子β1(transforming growth factor-β1,TGFβ1)及转化生长因子β1Ⅱ型受体(transforming growth factor-β1 type Ⅱ receptor,TβRⅡ)mRNA的表达。方法24只健康雄性Wistar大鼠,随机分为2组:模型组大鼠尾静脉注射二丁基二氯化物(dibutyltin dichlo-ride,DBTC)溶液7mg/kg,对照组仅注射相同体积的溶剂,28d时剖杀动物,采用HE染色和Masson染色观察两组大鼠胰腺组织病理变化,采用荧光定量PCR检测TGFβ1和TβRⅡ mRNA在胰腺组织的表达。结果模型组大鼠胰腺组织镜下可见纤维组织增生明显,胰腺小叶结构破坏或消失,腺泡萎缩;与对照组相比,模型组大鼠胰腺组织TGFβ1和TβRⅡ mRNA表达均升高(P<0.05)。结论TGFβ1和TβRⅡ在DBTC诱导的大鼠慢性胰腺炎胰腺组织表达升高。
Objective To detect the mRNA expression of transforming growth factor-β1(TGF-β1) and transforming growth factor-β1 type Ⅱ receptor(TβRⅡ) in pancreatic tissues of rats model with chronic pancreatitis.Methods Twenty-four healthy male Wistar rats were randomly divided into two groups:the control group and model group(n=10 per each group).Rat chronic pancreatitis model was induced by dibutyltin dichloride injection of(DBTC) into the right caudal vein with a syringe at a dose of 7 mg/kg body weight.The control group was treated only with solvent.Pancreatic tissues were collected after 28-day feeding,hematoxylin and eosin(HE) staining and masson trichrome staining were carried out to observe the pathological changes of pancrease,and the mRNA expression of TGF-β1 and TβRⅡ was detected using real-time PCR.Results The mRNA expression of TGF-β1 and TβRⅡ was all increased in model group compared with the control group(P0.05).Conclusion The mRNA expression of TGF-β1 and TβRⅡ is increased in pancreatic tissues of rat with chronic pancreatitis model induced with DBTC.
