摘要
研究转化生长因子βI型(TβRI)受体在人肝癌组织中的表达,探讨其临床意义。方法以125l-TGF-βI为配体的放射性配体结台分析法,检测了TβRI在30例肝癌组织及癌周肝组织和4例正常对照肝组织中的表达。分析了肝细胞癌组织中TβRI相对表达率与肝癌的一些临床病理资料的关系。结果肝癌组织TβRI表达较癌周肝组织及正常肝组织明显降低(P<0.01),后两者问TβRI表达一致。包膜侵犯/无包膜组和伴血管癌栓组TβRI表达率明显降低(P<005)。TβRI相对表达率与年龄、性别、HBV感染、肝硬化、AFP、肝癌的大小、数目及分化程度无关(P>005)。结论肝癌组织中TβRI表达降低可能是肝癌拮抗TGF-β对其生长抑制作用而恶性演进的关键因素,TβRI相对表达率与肝癌的预后有关。
Objective To study the expression of TβR I in HCC tissues and its clinical significance.Methods Expressions of TβR I in HCC tissues (HT, n=30) and surrounding liver tissues(HST, n=30) aswell as four normal control liver tissues (CIT, n=4) were determined by radioligand binding assay with125I TGFβ1 as the ligand. Relationships between some clinicopatholgical parameters of HCC and therelative expression rates of T6R I in HCC were studied. Results Compared with that in HST, theexpression of TβR I in HT was significantly lower than in HST and CIT (p<0.01). The expression ofTβR I in HST is similar to that in CTT. T6R I relative expression rates in the gfoup of no capsule orcapsule invaded and in the group of tumor embolus were markedly decreased (P<0.05). No closerelationships were found between the MR I relative expression rate and age, sex, HBV infection, livercirrhosis, AFP concentration, tumor diameter, tumor number, tumor differentiation as well as tumorembolus. Conclusions The low expression of TβR I in HT may be one of the key event which promotethe HCC malignant growth by protecting them against the growthinhibition effect of active TGFβ. TheTβR I relative expression rate may be related to the prognosis of HCC.
