摘要
目的:研究骨吸收抑制剂阿仑膦酸钠对骨质疏松模型种植体周围骨整合的影响。方法:27只5月龄日本大耳白兔随机分成3组,每组9只:单纯卵巢切除组(OVX组),伪手术组(S组),卵巢切除+阿仑膦酸钠组(ALN组)。其中OVX组和ALN组摘除双侧卵巢,而S组去除卵巢旁等量的脂肪组织。卵巢摘除后12周,双侧股骨远心端、胫骨近心端植入两枚种植钉,AL组腹腔给予Alendronate,OVX组给予等量的生理盐水,分别在4、8、12周处死动物,制作标本行扫描电镜观察,测定骨性结合率。结果:在种植钉植入4周后,3组的骨性结合率都未达到40%,在种植钉植入8周后,S组和ALN组的骨性结合率达到60%以上,而OVX组的骨性结合率仍然未达到50%,在种植钉植入12周后,ALN组的骨性结合率达到90%以上,S组的骨性结合率达到95%,OVX组的骨性结合率达到63%。结论:Alendronate的全身应用对骨质疏松兔的种植体周围的骨性结合率有着促进作用。
Objective: To investigate the influence of alendronate,a bone resoprtion inhibitor,on the osseointegration of implant-bone interface in estrogen-deficient rabbits.Methods: 27 five-month-old Japanese white female rabbits were randomly divided into three groups: an ovariectomy group(OVX),an ovariectomy+ Alendronate group(ALN) and a shamed-operated group(S).Rabbits of OVX group and ALN group had a surgical removal of bilateral ovaries,while rabbits of S group had a surgical removal of equivalent adipose tissue beside the ovaries.Two implants were installed into bilateral distal femurs and proximal tibias respectively 12 weeks after operation.Alendronate was administrated by intraperitoneal injection in ALN group;meanwhile equivalent normal saline was administrated in OVX group.After 4,8,12 weeks healing,both bilateral tibias and femurs were harvested.Specimens were prepared for the SEM observation to determine the rate of osseointegration.Results: None of the osseointegration rate in each group can reach 40% four weeks after implants installation.The osseointegration rate was beyond 60% in both S group and ALN group,while was below 50% in OVX group eight weeks after implants installation.Twelve weeks after implants installation,the osseointegration rates of the ALN group,S group and OVX group were 90%,95% and 63% respectively.Conclusion: Systemic application of Alendronate in osteoporosis rabbit models can improve the bone-implant osseointegration markedly.
