舌鳞状细胞癌的基因表达谱分析

摘要 目的:探讨舌鳞状细胞癌的候选基因。方法:采用包含有4096个cDNA克隆的基因芯片技术,分别对3例舌鳞状细胞癌组织及其相应正常组织进行基因表达谱的比较。结果:3例标本共同表达的差异表达基因共126条,其中Ratio>2的明显上调基因11条,而Ratio<0.5的明显下调基因6条。结论:生物信息分析显示:显著差异表达基因与肿瘤的发病机理存在相关性。 Objective: To study the candidate genes of tongue squamous cell carcinoma by the method of microarray.Methods: We study novel gene expression by cDNA microarray method.The total RNA from both 4 cases of tongue squamous cell carcinoma tissues and normal tongue squamous cell tissue were compared.The PCR product of 4096 genes was spotted onto a kind of chemical-material-coated-glass slide in array.Then they were reversely transcribed to cDNA with the incorporation of fluorescent-labeled dCTP to prepare the hybridization probes.After hybridization,cDNA microarray was scanned for the fluorescent intensity.TSCC-related gene expression was screened through the analysis of difference in gene expression profiles.Results: Among all target genes,there were 126 genes whose expression level differed from TSCC and normal tissues.After we analyzed the genes,we found that 17 genes were obviously related to TSCC.Conclusion: Our results suggest that significantly differential expression of genes may be associated with the pathogenesis and progression of the TSCC.
