圆斑星鲽肌钙蛋白Ⅰ基因的克隆和序列分析

Cloning and Characterization of Troponin Ⅰ gene from Verasper variegates

通过建立圆斑星鲽肌肉cDNA文库,大规模的EST测序和PCR技术得到了圆斑星鲽肌钙蛋白Ⅰ的完全表达序列,该序列全长845 bp,编码171个氨基酸。经同源性分析结果表明,与多种鱼类的肌钙蛋白Ⅰ具有较高同源性,其中与鳜鱼的肌钙蛋白Ⅰ同源性最高,为86%。序列提交GenBank收录,登录号为GU229275。

In this paper,the cDNA library from muscle of Verasper variegates was constructed using Creator SMART cDNA construction kit and large numbers of colonies were randomly picked and sequenced from the 5＇. One EST with high homology with Troponin I gene of other species was found and then the complete express sequence of troponin I from Verasper variegates was obtained by PCR. This gene was 845 bp with 516 bp of ORF,which encodes 171 amino acids and had the highest similarity with Siniperca chuatsi homologue with percent identities being 86% .
