摘要
为选育γ-癸内酯高产菌株,以毕赤酵母TT009(Pichia guilliermondiiTT009)为出发菌株进行原生质体紫外诱变,确定原生质体形成和诱变的最佳条件为菌龄16 h,酶解浓度1%,酶解时间50 min,酶解温度28℃,15 W紫外灯于30 cm处照射25 min。经初筛和复筛,得到γ-癸内酯高产菌株M6,利用该菌株进行摇瓶发酵,γ-癸内酯产量达到1.25 g/L,比出发菌株提高了28.8%。
Breeding on high yield of γ-decalactone produced by mutant of Pichia guilliermondii TT009 was studied by UV protoplast mutagenesis. This optimization of protoplast formation and regeneration were as following, 16 h of yeast incubation time, 1% of enzyme concentration, 28℃ of enzyme temperature. Results indicated that the optimization of UV protoplast mutagenesis was as following, 15 W UV light, 30 cm of lighting distance for 25 min. Through the preliminary and continual selection, a high yield γ- decalactone mutant M6 was obtained, which the yield of γ- decalactone was 1.25 g/L,improving the activity about 28.8% compared with TT009.
