摘要
目的:为了解感染日本血吸虫后小鼠不同脏器的免疫反应,我们观察了小鼠肝、脾、小肠Th2因子的变化并探讨其意义.方法:采用ABC免疫组化法,并用多媒体病理图文定量分析,动态研究感染后8,10,12周IL-4、IL-5、IL-10的变化.结果:在感染小鼠肝脏IL-4明显高于正常对照(P<0.01).并随感染时间的延长而逐步升高,IL-5和IL-10亦有相同变化,但IL-4最高,IL-10次之,IL-5居后;在感染小鼠的脾脏IL-4、IL-5、IL-10亦随感染时间的延长逐步缓慢升高,但IL-4在感染的第10周,IL-5在感染的第12周,IL-10从感染的第8周起与正常对照比较才有显著意义(P<0.05);在感染小鼠的小肠IL-4、IL-5、IL-10随感染时间延长呈缓慢升高趋势,从感染的第8周起与正常比较有显著差异(P<0.05~0.01).结论:感染日本血吸虫病的小鼠,肝脏是Th2因子免疫应答的主要器官,IL-4、IL-5和IL-10呈同步平行变化,以IL-4升高最明显;脾脏是调节免疫的器官,上述Th2因子的变化明显低于肝脏;而感染小鼠的小肠亦不是免疫应答的主要场所,其Th2因子的升高可能是整体反应的一部分.
Objective: To understand the changes of Th2 cytokines in livers, spleens and colons of miceinfected schistosomiasis japonica. Methods: ABC immunohistochemical stain technique and multimediapathologic pictures analpsis were used. Meanwhile, IL-4, IL-5 and IL-10 of infected mice werestudied in 8th, 10th and 12th weeks. Results: In the livers of infected mice, IL-4 was higher than thatof normal control.(P<0.01) lt was gradually increased with prolongated time. Also, the same changes forIL - 5 and IL -10 were observed. But the level of IL-4 was the highest in three cytokines. IL- 10 wassecond and IL- 3 third. In the spleens of infected mice, IL-4, IL-5 and IL- 10 were gradually cle-vated. However, they were slower than those of livers. IL-4 increased in 10th week, IL-5 in 12thweek and IL-10 in 8th week, respectively.There were statistical significance. (P<0.05) In the colonsof infected mice, IL-4, IL-5 and IL-10 slowly increased with prolongated time. There were statisticaldifference, too. (P<0.05~0.01) Conclusions: In mice infected schistosomiasis japonica, the immuneresponse happened mainly in the liver. All of IL-4, IL- 5 and IL- 10 were obviously increased. Thespleen played an immune regulation role. The level of Th2 cytokines was lower than that of livers.As for colons, it was not the main organ of immune reaction. The increase of Th2 Cytokines only representeda part of immune reaction in the body.
