摘要 采用mRNA差异显示(DDRT-PCR)技术,分离枇杷‘早钟6号’幼果在0℃低温胁迫6h后差异表达的基因。共分离到19个差异表达的基因片段,经反向Northern杂交,得到了4个阳性的枇杷低温诱导相关基因片段(编号为CSIGE1~4-Cold Stress Induced Genes in Eriobotrya japonica1~4)。结果显示:CSIGE1只在低温处理的枇杷果实中表达,这表明该基因与低温逆境有关;CSIGE2和CSIGE3是受低温诱导的上调表达基因;而CSIGE4登录号为GT617706,未搜索到与任何基因有相似性,可能为新基因。 The DDRT-PCR technique was used to analyze genes differential expression in the fruit of loquat cultivar‘Zaozhong 6’treated at 0 ℃ for 6 h. Nineteen differentially expressed gene segments induced under cold stress were obtained, four of which were confirmed as positive clones by Reverse Northern dot blotting and named CSIGE 1~4-Cold Stress Induced Genes in Eriobotrya japonica 1-4. The results showed that CSIGE 1 was only expressed in loquat fruit treated at the low temperature, indicating that this gene was related to cold acclimation. CSIGE 2 and CSIGE 3 were up-regulated genes induced under cold stress, and CSIGE 4 (Log No. GT617706) did not show homology to any gene, which may be a new gene.
