摘要
目的:探讨不同经脉配穴法选穴针刺对急性胃黏膜损伤大鼠相关细胞因子表达的影响。方法:SD大鼠60只,10只为空白组;50只制作为急性胃黏膜损伤模型,随机分为模型组10只和针刺组A、B、C、D各10只。针刺组A取本经配穴,B取表里经配穴,C取同名经配穴,D取他经配穴,分别给予电针治疗7d。7d后放射免疫法测定各组胃黏膜及血浆PGE2、SS、TNF-α的浓度;免疫组化法检测胃黏膜EGF和TGF-α的表达。结果:PGE2浓度,针刺组A明显高于模型组和针刺组D;胃黏膜及血浆SS浓度,针刺组A、B、C均显著高于模型组,且A明显高于B、C、D;胃黏膜及血浆TNF-α浓度,针刺组A、B、C均显著低于模型组、针刺组D(P<0.01),其中A、B低于C;与其它组比较,针刺组A能上调EGF及TGF-α的表达(P<0.05,0.01)。针刺组B能上调EGF的表达(均P<0.05,0.01)。结论:通过选穴针刺对大鼠急性胃黏膜损伤修复的作用机制可能与抑制炎性介质TNF-α释放,促进内源性保护物质PGE2、SS合成及EGF和TGF-α在胃黏膜的表达有关,但存在差异。本经配穴法作用最明显,其保护效应与以上各方面均相关;表里经配穴法侧重于调整TNF-α、SS的合成、释放及促进EGF的表达;同名经配穴法侧重于调整TNF-α、SS的合成、释放;他经配穴法则对上述调整作用不明显。
Objective:To observe the effect of different acupoint combination on cytokine of acute gastric mucosal lesion rats.Methods:Sixty SD rats were divided randomly into 6 groups,10 rats each group.Group A was blank control group.Group B was model group.In group C,combining points were used on the same meridians.group D,combining points were used on the exteriorly-interiorly related meridians.In group E combining points were used on the meridians sharing the same name.In group F,combining points were used on the unrelated meridians.Acute gastric mucosal lesion rat model was built by ethanol ingestion.Radioimmunoassay was used to determine contents of PGE2,SS,TNF-α,and immunohistochemistry for expression of EGF and TGF-α.Results:As compared with groups B and F,content of PGE2 was increased in group C.As compared with group B,the content of SS was obviously increased in groups C,D and E,and that in group C was obviously higher than in groups D,E and F.As compared with groups B and F,TNF-α was decreased in groups C,D and E(P0.01),and that in groups C and D was obviously lower than in group E.As compared with other 5 groups,TGF-α and EGF expression levels were increased obviously in group C.All P0.05 or P0.01.Conclusion:The reparation effect of acupoints of the same meridians might be to inhibit TNF-α release,promote PGE2 and SS synthesis,and promote the expression of EGF and TNF-α in gastric mucosa.The reparation effect of acupoints of the exteriorly-interiorly related meridians might be to regulate TNF-α and SS synthesis,promote the expression of EGF.The reparation effect of acupoints of the meridians sharing the same name might be to regulate TNF-α and SS synthesis and release.
