苦参素抗大鼠肝脏缺血再灌注损伤作用的研究

摘要 目的探讨苦参素注射液对大鼠缺血再灌注肝脏细胞Bax、Bcl-2及Caspase-3蛋白表达的影响及它们与肝脏细胞凋亡的内在联系。方法健康SD大鼠30只随机分为3组,空白对照组(S组)、缺血再灌注+生理盐水组(I/R组)、缺血再灌注+苦参素注射液组(M组),每组10只。免疫组化检测Bax、Bcl-2及Caspase-3蛋白的表达,每组选24个视野分别测量光密度值(OD值)。结果 I/R组BaxOD值明显高于S组(P<0.01),SF组BaxOD值明显低于I/R组(P<0.01),且低于S组(P<0.05)。I/R组Bcl-2OD值高于S组(P<0.05),且SF组Bcl-2OD值高于S组(P<0.01),但I/R组与SF组比较差异无显著性。I/R组Caspase-3OD值明显高于S组(P<0.01),且明显高于SF组(P<0.01)。结论丹参注射液增加肝脏Bcl-2蛋白的表达,降低Bax及Caspase-3蛋白的表达,抑制肝脏细胞凋亡,保护缺血再灌注肝脏。 Objective To explore the effects of matrine on the expression of apoptosis proteins,including Bax,Bcl-2 and Caspase-3 following acute liver ischemic reperfusion in rats.Methods 24 SD rats were randomized into three groups(n=8 each):sham group(S group),ischemic reperfusion +NS group(I/R group),ischemic reperfusion+matrine(M group).The exp ressions of Bax,Bcl-2 and Fas were detected by immunohistochemistry.Results The average OD value of Bax was signif icantly higher in I/R group than in S group(P0.01),but was signif icantly lower in M group than in I/R group(P0.01),or than that in S group(P0.05).Compared with S group and M group,the average OD value of Bcl-2 was higher and signif icantly higher(P0.05,P0.01),and the average OD value of Caspase-3 was signif icantly higher in I/R group than in S and M group(P0.01).Conclusion Matrine can enhance the expression of Bcl-2,decrease the expression of Bax and Caspase-3,inhibit liver cell apoptosis and protect liver injury after ischemic reperfusion.
