单克隆抗体研制中杂交瘤细胞的冻存与复苏

Frozen Storage and Resuscitation of Hybrid Tumour Cells in Manufactyring Monoclonal Antibody

为寻找出简单有效的杂交瘤细胞冻存与复苏方法，试验以３组不同组分的细胞冻存液，用３种方法冻存杂交瘤细胞。结果表明：使用Ⅲ号液（５０％小牛血清＋１０％二甲基亚砜＋４０％ＲＰＭＩ－１６４０）采用方法Ⅲ（液氮面６～１０ｃｍ存放３０ｍｉｎ后直接浸入液氮中），细胞冻存与复苏效果最佳。

In order to find a simple and effective approach for frozen storage and resuscitation of bybrid tumour cells 3 groups of cells frozen storage solutions with different components and 3 different storing approaches were used respectiely in the test. The results show that the best effect for cells frozen storage and resuscitation was obtained by a combination of the solution no.3(50% calf serum + 10%dimethyl sulfoxide + 40%RPMI-1640) and approach no.3 (Storing cells at 6～10cm below liquid nitrogen surface for 30 minutes and then directly putting them into liquid nitrogen).
