摘要
应用SDS-聚丙烯酰胺凝胶电泳(PAGE)、琼脂免疫扩散和免疫电泳分析比较了鲤形目鲤亚目的鲫与其他8种淡水鱼的血清、体表粘液蛋白及其抗原相关性。这8种鱼分别是鲤亚目鲤形科的鲤、鳊、草鱼,鳅科的泥鳅,鲇亚目鲇科的鲇,鱼尝科的黄颡鱼,鳢形目乌鳢科的乌鳢和合鳃目合鳃科的黄鳝等。结果表明,在SDS-PAGE图谱中各种鱼的血清和体表粘液共有和特有蛋白组分的比例数在不同目、科和种之间差别不明显。在琼扩图谱中,兔源抗鲫血清蛋白抗体与鲫血清和体表粘液蛋白产生2~3条沉淀带;在免疫电泳图谱中产生10条以上沉淀带。该抗体不能与乌鳢和黄鳝的血清和粘液蛋白结合,但能与同目的其他6种鱼的血清和体表粘液蛋白产生不同数量的沉淀带,其数目、部位和密度与其亲缘属性相一致。此外,同种鱼类的血清和体表粘液抗原表现出明显的相关性。所有这些表明,不同鱼种的血清和体表粘液蛋白分子存在与亲缘程度相应的交叉抗原。
The relationship of molecules and cross antigens of the serum and body surface slime proteins among Carassius auratus and eight otheir fish species under three orders and five families was analysed by SDS-PAGE,gel diffusion and immunoelectrophoresis.The bands of the serum and body surface slime proteins of the tested species in the map of SDS-PAGE showed no significant difference.The antibodies to C.auratus' serum proteins could react with both its serum and its body surface slime proteins,producing three and two precipitation bands respectively in the gel diffusion,at least ten precipitation bands in the immunoelectrophoresis.In addition,except those of O.argus and M.albus,the antibodies could bind those of other six species.These results showed that there existed the cross antigens in serum and body surface slime among various fish species,which corresponded with their relationship to each other.
