Curcumin-induced apoptosis of human liver cancer Bel-7402 cell line

摘要 目的探讨姜黄素对人肝癌Bel-7402细胞的增殖抑制、诱导凋亡及其分子作用机制。方法用姜黄素10μmol/L体外处理Bel-7402细胞48h。采用MTT比色法检测细胞生长的抑制作用,流式细胞术检测细胞凋亡率,RT-PCR检测Bcl-2和BaxmRNA表达。结果10μmol/L姜黄素处理细胞48h后呈现时间和剂量依赖性细胞增殖抑制作用;细胞凋亡率随着姜黄素浓度的增加而逐步增高,BaxmRNA表达上调,而Bcl-2mRNA表达降低。结论姜黄素对Bel-7402细胞有显著的增殖抑制及诱导凋亡作用,其作用机制可能与上调Bax基因的表达及下调Bcl-2基因的表达有关。 Objective To investigate the apoptosis of human liver cancer Bel-7402 cell line induced by curcumin in vetro.Methods The cells were treated in vetro with curcumin 10μmol/L for 48h.Inhibiting effect of cell proliferation was detected with MTT assay,cell apoptosis was analyzed by flowcytometry,and the expressions of Bcl-2 and Bax mRNA were detected by RT-PCR.ResultsAfter treated by curcumin for 48 h,the cell proliferation was inhibited in time-and dose-dependent manner.As the concentration of curcumin increased,the apoptosis rate of the cells increased,expression of Bax mRNA up-regulated and that of Bcl-2 mRNA down-regulated.ConclusionCurcumin inhibits cell proliferation and enhances cell apoptosis,which may be related to up-regulating Bax mRNA expression and down-regulating Bcl-2 mRNA expression.
