Objective: To evaluate the immunological contraceptive effects and the cytotoxicity induced by recombinant protein vaccine of mouse sperm Sp17 fused with IL-5 (r-Sp17-IL-5). Methods: The female mice were immunized with r-Sp17-IL-5 protein vaccine by inate. Immunocontraceptive effects of r-Sp17-IL-5 were detected, including pathologic changes of vital organs and changes in the rates of teratism and micronucleus of sperms in filial generation. Results: Following intranasal immunization, the pregnancy rates of the Sp17-IL-5, IL-5+Sp17 and Sp17 groups were decreased to 23.1%, 28.6%, 57.1%, respectively. The number of newborn per birth in each group was decreased too. There were no apparent pathologic changes in the liver, spleen, brain, kidney and lung. Micronucleus tests of bone marrow obtained from immunized mice and their offspring showed that there was no significant difference in polychromatic erythrocyte/normochromatic erythrocyte (PCE/NCE) and micronucleus rate between the experimental and normal groups. Conclusion: The recombinant protein vaccine of mouse sperm Sp17-IL-5 has a stable reduction in fertility, and can not produce the cytotoxicity to the immunized mice and their offspring.
