实验性小鼠肺纤维化过程中IL-31mRNA的动态表达及意义

摘要 目的:探讨博莱霉素(BLM)致肺纤维化小鼠各时期肺组织中白细胞介素31(IL-31)mRNA的表达规律及其意义。方法:将48只雄性昆明种小鼠随机分为对照组(n=24)和实验组(n=24)。分别给予生理盐水和博莱霉素(5mg/kg)一次性气管内灌注,于灌注第7、14、21和28天处死小鼠,取肺组织,应用RT-PCR法测定其中的IL-31mRNA的相对转录水平。结果:1实验组肺组织中IL-31mRNA表达水平在第7、14、21天均高于对照组(P<0.05)。2实验组中IL-31mRNA表达从第7天开始升高,第21天达到高峰。结论:IL-31在肺纤维化形成过程中存在高表达,尤其在介导中后期肺纤维化进程中发挥重要作用。 Objective:To investigate the IL-31mRNA expressions in lung tissues of experimental mice with pulmonary fibrosis(PF)induced by bleomycin in various periods and to study the effects of IL-31 in pulmonary fibrosis.Methods:Forty eight male mice were randomly divided into two groups :control group(n=24)and experimental group(n=24),and poured respectively by saline and bleomycin solutions.They were sacrificed on the 7th,14th,21th and 28th day.RT-PCR method was used to analyse the mRNA expression of IL-31.Results:① The mRNA expression of IL-31 in lung tissues in experimental group were higher than those in control group(P0.05).② The mRNA in experimental group were higher than those in control group from the 7th day,peaked on the 21th day.Conclusion:The expression of IL-31 was high at different stages of pulmonary fibrosis.IL-31 played an important role especially at the middle and late stages in pulmonary fibrosis.
