摘要
为了探讨显微操作组织贴块法培养大鼠动脉血管内皮细胞的效果,试验用倒置相差显微镜观察细胞形态和生长特征,用免疫细胞化学法检验内皮细胞Ⅷ因子进行血管内皮细胞鉴定。结果表明:72h后内皮细胞从组织块边缘游出,12d后细胞开始融合成片,内皮细胞阳性率为97%。说明应用显微操作组织贴块法培养能获得高纯度血管内皮细胞。
To explore effects of micrurgy and tissue explants adhension to culture vascular endothelial cells from albino rat aorta.The cell growth was evidenced by revealing the morphological characteristics under inverted phase contrast microscope and detection of Ⅷ factor(or von Willebrand factor,vWF)by immunocytochemistry.The results showed that the migration of endothelial cells from aortic cub was found after 72h's cultivation,forming confluent monolayer of a cobblestone pattern after 12 days.Cell positive rate of vWF in the cytoplasm of the new cells was 97%.Combination micrurgy with tissue explants adhension could obtain pure vascular endothelial cells.
