The effect of immunoinflammatory cytokines on human fetal pancreatic islet function

摘要 目的研究炎性细胞因子刺激对人胚胎胰岛分泌功能的影响。方法采用常规消化法分离人胚胎胰岛并进行原代培养,分别加入rhTNFα、rhIL-1β、rhIFNγ或同时加入2种或3种细胞因子,培养24和48小时取其上清测定胰岛素、胰升糖素含量。结果3种细胞因子单独或联合应用均可不同程度地抑制胰岛素的分泌,联合应用对抑制胰岛素分泌有协同作用;对胰升精素的分泌则有明显的促进作用,但联合应用的协同作用因胰岛培养时间的不同而异。结论细胞团子影响胰岛细胞分泌功能的机制可能是复杂多样的。 Objective The aim of this investigation was to study the effects of immunoinflammatory cytokines onfetal islet secretory function in vitro. Methods The islets from fetal pancreas were separated by collagenase type Ⅴ(0. 5 mg/ml) and cultured in vitro. The islets were exposed to culture medium alone or with different kindsinflammatory cytokines for 24 or 48 h. The isupernatants from culture of human fetal islets were assayed for theconcentrations of insulin and glucagon. Results Insulin secretion in all groups was inhibited to different degrees afterislets were exposed to inflammatory cytokine (s) alone or in different combinations. There was a synergistic inhibitingeffect on insulin secretion after exposure to 3 kinds of cytokines at the same time. On the other hand, glucagon secretionin all groups was significantly increased with synergistic promoting effect after exposure to the immunoinflammatorycytokines. Conclusion The secretory function of pancreatic islets was affected dose-and time-dependently byimmunoinflammatory cytokines.
