摘要
目的:研究小鼠心肌细胞上七种Mg2+转运体mRNA在不同发育时期的表达情况。方法:提取不同发育时期小鼠心脏组织mRNA,用七对Mg2+转运体特异性引物进行RT-PCR扩增,经凝胶成像仪进行凝胶荧光定量检测。结果:经RT-PCR半定量检测后,观察到小鼠心肌细胞上七种转运体mRNA在不同时期都有不同程度的表达,其中TRPM7和MagT1两种转运蛋白在各个时期表达较稳定,并且表达量明显高于其他五种Mg2+转运体。结论:TRPM7和MagT1在小鼠心肌细胞不同发育时期Mg2+调控过程中可能发挥主导作用。
Objective:To investigate the expression levels of seven Mg2+ transporters in mouse cadiocytes of different age.Methods:The mRNA of different age mouse cadiocytes was abstracted and the mRNA of seven Mg2+ transporters was amplified by semi-quantitative RT-PCR with specific primers.The fluorescence intensity was detected by gel formatter.Results:All seven Mg2+ transporters were detected at different ages from new born to 60-day after birth,although the expression levels of TRPM7 and MagT1 showed relatively stable and higher than the others.Conclusion:TRPM7 and MagT1 might play important roles in regulating Mg2+ in mouse cardicytes.
