摘要
目的研究体外培养大鼠脑皮质星形胶质细胞不同液压冲击损伤下形态学改变。方法根据Scott’s细胞液压冲击损伤原理,建立体外培养星形胶质细胞液压冲击损伤模型。原代培养SD大鼠脑皮质星形胶质细胞,分为对照组、0.05MPa、0.10MPa、0.20MPa、0.40MPa、0.60MPa冲击压力组(n=8),观察细胞形态学变化和培养上清LDH含量变化。结果星形胶质细胞平均坏死率随着压力增高而显著增高,当压力达到0.6MPa时,细胞的平均坏死率达到51.11%。0.20MPa组培养上清LDH含量与对照组相比升高显著,当压力达到0.40MPa以上时培养上清LDH含量与对照组相比反而成下降趋势。结论0.20MPa液压冲击压力比较适合进行星形胶质细胞的损伤性研究。
Objective To observe and study the rat cerebral cortical astrocytes (AS) in different morphological changes in vitro after fluid percussion injury. Methods According to Scott's principle of cell hydraulic impact injury; set up the astrocytes in vitro fluid percussion injury model. SD primary cultured rat brain conical astrocytes were divided into control group, 0.05MPa, 0.10MPa, 0.20MPa, 0.40MPa, 0.60MPa impact pressure group (n = 8), and observe the morphology changes and cultivation lactate dehydrogenase (LDH) content in the culture supernatant. Results The average necrosis rate of astrocytes increased significantly as the pressure increased. When the pressure reached 0.6MPa, the average necrosis rate is 51.11%. Comparing with the control group, 0.20MPa group LDH content of culture supernatant increased significantly. Increasing the pressure (0.40MPa and above), LDH content of culture supernatant, as compared with the control group, trended into a downward. Conclusion 0.20MPa pressure of fluid percussion injury is more suitable for AS research.
