大鼠脊髓高能量冲击伤后细胞凋亡的研究

摘要 目的本实验通过研究大鼠急性脊髓损伤后神经细胞凋亡,探讨急性脊髓损伤后神经细胞凋亡的时空分布特点。方法 Wistar成年健康大鼠46只,随机分为三组。垂直打击该段脊髓背侧正中制作急性脊髓损伤模型。于SCI后,8h,24h,7d,14d及21d,采用苏木素一伊红染色(HE)及凋亡细胞原位缺口末端标记(TUNEL)法对急性损伤的脊髓组织进行检测。结果 1)损伤组凋亡细胞24h达到高峰然后明显减少,各时间段差异明显(P<0.05),21d仍可见少量凋亡细胞。2)损伤组中10g组与20g组在各时间段有明显差异(P<0.05)。结论高能损伤后细胞凋亡时间相的分布特点;随着损伤的加重,损伤组各组随时间的延长凋亡细胞逐渐增多,术后24h达高峰。 Objective To investigate the cellular distribution of apoptosis following T11-T12 spinal cord injury. Methods 46 Wistar rats with a body weight used in these experiments fell into three groups randomly. Tarlov behaviour score was performed before surgery and after surgery at 8 hours, 3 days, 7days, 14days, 21days(n= 3for each time point). After injury, TUNEL--positive cells were found in the compression region. Results After injury, TUNEL--positive cells were found in the compression region after8h. Conclusions With the aggravation of stretch injury, the number of TUNEL-- positive cells increased and reached a summit 24h after injury and then decreased gradually from7days to 21days after injury.
