摘要
目的研究藻蓝蛋白对2型糖尿病大鼠肝脏和胰岛形态结构和细胞凋亡的影响。方法健康雄性Wistar大鼠32只,高糖高脂饲料喂养联合小剂量链脲佐菌素(STZ)腹腔注射建立2型糖尿病大鼠模型,藻蓝蛋白灌胃治疗,组织病理学技术观察肝脏和胰岛细胞的形态结构,用原位末端标记法(Tunel法)检测肝细胞和胰岛细胞凋亡。结果模型组大鼠肝组织结构紊乱,中央静脉扩张,肝细胞肿胀,颗粒变性和脂肪变性;胰岛萎缩、结构模糊,细胞数目减少、排列紊乱,胞浆肿胀、空泡变性等;两个组织中凋亡阳性细胞较正常组明显增多(P<0.05);藻蓝蛋白组肝脏组织和胰岛细胞形态结构较清晰,正常细胞数目增多、排列较规则,肿胀和变性细胞较少,凋亡阳性细胞较模型组显著减少(P<0.05)。结论藻蓝蛋白可抑制糖尿病大鼠肝脏和胰岛细胞凋亡,发挥一定的保护作用。
Objective To investigate the effect of phyeocyanin on the morphosis and apoptosis of liver and islet cells in rats with type 2 diabetic disease. Methods Type 2 diabetes mellitus models were induced by high sugar and fat diet and low dose intraperitoneal injection of streptozotocin (STZ) in 32 healthy male Wistar rats, and treated with phycocyanin by intragastrie administration. The morphosis of liver and islet cells were observed by histopathological technique and the apoptosis of liver and islet cells were detected by Tunel assay. Results In the model group, the hepatic tissue structure showed disorderly and central vein extension and the islet shrink and ambiguity, the number of islet cells reduced and arranged disorderly with plasma swelling and vacular degeneration, and the number of positive apoptotic cells and apoptosis index were significantly higher than those of two tissues in normal group (P〈0. 05). In phycocyanin group, the morphosis of liver and islet cells improved significantly and the number of nor mal cells increased regularly with decreasing of swelling and degenerative cells, and the number of posi tive apoptotic cells and apoptosis index were significantly lower than those in model group (P〈0. 05). Conclusion Phycocyanin might play a protective effect by inhibiting apoptosis of lver and islet cells in di abetes mellitus rats.
