还原型谷胱甘肽对肝缺血再灌注损伤细胞凋亡的影响 被引量:6

摘要 目的探讨还原型谷胱甘肽对家兔肝缺血再灌注损伤肝细胞凋亡的影响及其机制。方法将90只健康家兔随机分为对照组、肝缺血再灌注组和还原型谷胱甘肽保护组,制备肝缺血再灌注损伤模型。免疫组化法检测Bcl-2和Bax表达,原位细胞凋亡法检测肝细胞凋亡。结果与正常肝组织比,肝缺血再灌注损伤时,Bcl-2蛋白表达减少,Bax表达增高,细胞凋亡指数增加。使用还原型谷胱甘肽后,上述指标的异常变化均明显减轻,其有显著性差异(P均<0.05)。结论还原型谷胱甘肽通过上调Bcl-2蛋白的表达,抑制Bax蛋白的表达,从而抑制肝细胞凋亡。 Objective It is to approach the influence of reduced glutathione on the apoptosis of hepatocytes in rabbits with hepatic ischemia reperfusion injury.Methods 90 healthy rabbits were randomly divided into control group,ischemia reperfusion group and reduced glutathione protection group.The model of hepatic ischemia reperfusion injury was copied.The expression of Bcl-2 and Bax were measured with immunohisto-chemical method.Cell apoptosis were observed by TUNEL.Results The expression of Bcl-2 protein was significantly decreased in ischemia reperfusion group than that in control group(P〈0.05).The expression of Bax protein and apoptosis indexes were significantly increased in ischemia reperfusion group than that in control group(P〈0.05).After treatment with reduced glutathione,the abnormal changes of all above parameters were diminished remarkably(P〈0.05).Conclusion Reduced glutathione may decrease the apoptosis of hepatocytes through increasing Bcl-2 expression and inhibiting Bax expression.
