摘要
目的观察早期自然流产患者绒毛组织中RECK基因(基质金属蛋白酶(MMP)抑制基因)的表达及其与MMP-2、-9之间的关系。方法采用半定量RT-PCR和Western blotting法检测20例早期自然流产患者和20例正常早期妊娠绒毛组织中RECK、MMP-2和MMP-9基因的表达情况。结果①早期自然流产患者绒毛组织中MMP-2及MMP-9的mRNA表达水平低于正常早期妊娠者,差异有统计学意义(P<0.05);RECKmRNA的表达高于正常早期妊娠者,差异有统计学意义(P<0.05)。②自然流产患者绒毛组织中MMP-2、MMP-9蛋白表达低于正常早期妊娠者,差异有统计学意义(P<0.05);RECK蛋白表达高于正常早期妊娠者,差异有统计学意义(P<0.05)。③RECK蛋白与MMP-2、MMP-9蛋白之间存在明显的负相关。结论RECK表达的增加进而抑制MMP-2、MMP-9的活化,致使滋养细胞侵袭能力下降,可能与孕早期自然流产的发生有关。
Objective To investigate the expression of matrix metalloproteinase suppressor gene RECK and its relationship with matrix metalloproteinase-9(MMP-9) and MMP-2 in villi in patients with early spontaneous abortion(ESA).Methods The expression of RECK,mRNA and protein of matrix metalloproteinases MMP-9 and MMP-2 in villi of ESA patients(abortion group,n=20) and normal first trimester pregnant women(control group,n=20) was detected by semi-quantitative RT-PCR,Western blotting,respectively.The correlations between the expression of RECK and the expression of MMP-2,MMP-9 were analyzed by Pearson test.Results ①The expression of MMP-9 and MMP-2 mRNA was significantly lower in villi of the abortion group compared with that of the control group(P0.05).The expression of RECK mRNA increased significantly in villi of the abortion group(P0.05);②Western blotting analysis demonstrated that the expression of MMP-9 and MMP-2 protein in villi of the abortion group was significantly lower than that of the control group(P0.05).The expression of RECK protein in villi was significantly higher in the abortion group than in the normal control group(P0.05);③The expression of RECK protein in villi of first trimester had significant negative-correlation with that of MMP-2 and MMP-9.Conclusion The increase in the expression level of RECK inhibited the activation of MMP-9 and MMP-2,and resulted in a decrease in the trophoblastic invasion of human placenta,which might be correlated with occurrence of first trimester spontaneous abortion.
