摘要
目的探讨二烯丙基二硫(DADS)对人结肠癌SW480细胞裸鼠致瘤的影响。方法建立人结肠癌裸鼠移植瘤模型。15只实验裸鼠分为3组,每组5只。观察DADS对裸鼠致瘤的影响,并记录各组裸鼠及其皮下移植瘤的生长情况,绘制肿瘤生长曲线,计算肿瘤体积和肿瘤生长抑制率。光镜下观察移植瘤组织形态学改变,采用免疫组织化学法检测瘤组织内增殖细胞核抗原(PCNA)、p21WAF1蛋白表达情况,应用流式细胞术检测移植瘤细胞周期分布。结果 DADS能明显降低人结肠癌SW480细胞致瘤性,抑制移植瘤生长,降低肿瘤组织异型性,引起细胞周期G2/M阻滞,抑制PCNA蛋白表达,增强p21WAF1蛋白表达。结论 DADS可明显降低人结肠癌SW480细胞的致瘤性,抑制SW480细胞增殖,引起G2/M期阻滞,其可能与抑制PCNA蛋白表达、增强p21WAF1蛋白表达有关。
Objective To investigate the effects of DADS on the tumorigenesis of the human colon cancer SW480 cell line in nude mice.Methods Fifteen nude mice which were used to establish xenograft tumor model were randomly divided into 3 groups.The effects of DADS on the tumorigenesis were observed,which included the growth of nude mice,the tumor growth curve and inhibition rate.The morphology of tumor was observed under optical microscope.The expression of proliferating cell nuclear antigen(PCNA),p21WAF1 was detected by immunohistochemistry and morphometric quantitative analysis.Cell cycle distribution was analyzed by flow cytometry(FCM).Results The tumorigenesis of the human colon cancer SW480 cell line in nude mice was inhibited markedly,and the growth of xenograft tumor also were inhibited significantly by DADS.After treated by DADS,The cellular atypia and nuclear-cytoplasmic ratio were decreased,cell cycle were arrested in G2/M,the expression of PCNA were suppressed,while the expression of p21WAF1 were enhanced.Conclusion DADS could decreae the tumorigenesis and inhibit the growth of the human colon cancer SW480 cell line in nude mice,arrested cell cycle in G2/M,which maybe relate to downregulated expression of PCNA and upregulated expression of p21WAF1.
