摘要
为了从母系遗传角度深入阐明西畴乌骨鸡的群体遗传背景,采用PCR直接测序法测定了36只西畴乌骨鸡的线粒体DNA D-loop区第I高变区520 bp序列。结果表明:在所分析的西畴乌骨鸡D-loop区序列中,T,C,A和G平均含量分别为30.3%,29.7%,27.4%和12.6%;共检测到核苷酸变异位点26个,核苷酸多样度为0.014 52±0.000 98;D-loop区序列存在12种单倍型,单倍型多样度为0.884±0.027,表明西畴乌骨鸡遗传多样性较丰富。聚类分析显示西畴乌骨鸡聚为A,B两大支,其中,A世系分为3小支,B世系分为2小支,说明西畴乌骨鸡在母系血统上存在较大遗传分化。
In the present study,36 Xichou black-bone chickens were selected from Xichou County,Yunnan province.The 520 bp HVS-I region of mitochondrial DNA D-loop was determined by direct sequencing of PCR products.The results revealed that the average percentage of T,C,A and G were 30.3%,29.7%,27.4% and 12.6%,respectively.Totally,26 mutation sites and 12 haplotypes were detected.The nucleotide diversity was 0.01452±0.00098,and the haplotype diversity was 0.884±0.027.Phylogenetic analysis by NJ method showed that all the sequences were clustered in 2 major clades: A clade which was further divided into 3 subclades and B clade which was further divided into 2 subclades.The results in this study indicated that there was remarkably genetic differentiation for the maternal origin of Xichou black-bone chicken.
