摘要
采用纤维素酶从脱脂脱糖的苹果渣中提取可溶性膳食纤维。通过正交试验优化,确定最佳提取条件为:纤维素酶添加量为4%,温度为50℃,料液比为1:15,酶解时间为6h。在该条件下,可溶性膳食纤维提取率可达142.34%。通过凝胶色谱(GPC)分析可知,可溶性膳食纤维主要为相对分子质量4523g/mol的低聚糖且分子量分布较窄,同时还有部分相对分子质量为4.417×105g/mol的大分子多糖,其分子量分布较宽。
Using cellulase to degrate the apple pomace which is eliminated fat and free-sugars.The orthogonal test was adopted to explore the best technological conditions of enzymolysis process.It was found that,for the extraction under the best addition of cellulose,temperature,ratio of solid to liquid and extraction time were 4 %,50 ℃,1:15 and 6 h respectively.The extraction rate of soluble dietary fiber was 142.34 %.Determination of the molecular distribution of apple pomace's SDF by GPC showed its average molecular weight was 4 523 g/mol.Its molecular weight distributions was narrow and may contain mostly oligosaccharide,while the other was about 4.417×105 g/mol comprising macromolecular polysaccharide which molecular weight distribution was wider.
