摘要
研究以风信子Gipsy queen的鳞片及幼叶为外植体直接诱导不定芽生成,结果表明:Gipsy queen单鳞片、双鳞片的初代最佳诱导培养基为:MS+6-BA5.0mg·L-1+NAA0.1mg·L-1,且双鳞片诱导效果好于单鳞片,诱导率达100%,增殖系数达10;幼叶外植体初代最佳诱导培养基为:MS+6-BA3.0mg·L-1+NAA0.2mg·L-1,幼叶作为外植体比鳞片诱导效果好,诱导率达100%,增殖系数达26.27;最佳继代培养基为:MS+6-BA2.0mg·L-1+NAA0.5mg·L-1+KT0.2mg·L-1,增殖系数达2.91;生根诱导最佳培养基为:1/2MS+NAA0.2mg·L-1,生根率可达93.33%。
Adventitious shoots were induced from scale segments and tender leafs of Hyacinthus orientalis L.cv.Gipsy Queen.As the result of this experiment: the appropriate medium for initial culture of single-scale and twin-scale of Gipsy Queen was: MS+6-BA5.0mg·L^-1+NAA0.1mg·L^-1,and twin-scale was better than single-scale.The induction rate was 100%,the proliferation multiple was 10.The appropriate medium for tender leafs was: MS+6-BA3.0mg·L^-1+ NAA0.2mg.L^-1,and the effect was better than that of scale segments.The induction rate was 100%,the proliferation multiple was 26.27.The best medium on subculture multiplication was MS +6-BA2.0mg·L^-1 +NAA0.5mg·L^-1 +KT0.2mg·L^-1.The proliferation multiple was 2.91.The ratio of root induction for the bulblets was 93.33% on the medium of 1/2MS+NAA0.2mg·L^-1.
