摘要
目的研究不同吸烟量对大鼠肺组织中核因子κB(NF-κB)和E-选择素表达水平的影响,探讨吸烟与COPD肺组织炎症的关系。方法雄性Wistar大鼠24只,随机分为对照组、小量吸烟组和大量吸烟组,每组8只。吸烟时间为20周。采用免疫组化和原位杂交半定量检测各组支气管上皮细胞NF-κB和E-选择素的表达。结果小量吸烟组、大量吸烟组NF-κB与E-选择素mRNA及蛋白表达均较对照组显著增高,小量吸烟组较大量吸烟组表达降低。E-选择素mRNA和蛋白的表达水平与NF-κB mRNA和蛋白的表达水平呈正相关。结论吸烟可引起大鼠气道上皮细胞NF-κB和血管内皮细胞E-选择素mRNA及其蛋白的表达水平升高,且随着吸烟量的增加其表达水平增高。
Objective To explore the effect of different smoking quantity on NF-κB and E-selectin expression,and the relationship of different smoking quantity with inflammation of lung tissues in chronic obstructive pulmonary disease(COPD). Methods Twenty-four Wistar rats were randomly divided into control group,low-dose smoking group and high-dose smoking group. The expression of NF-κB and E-selectin in rat lung tissues was detected by immunohistochemistry and hybridization in situ. Results The expression of NF-κB mRNA and protein in bronchial endothelial cells was significantly higher in high-dose smoking group and low-dose smoking group than in control group,and also higher in high-dose smoking group than in low-dose smoking group. The expression of E-selectin mRNA and protein showed the similar pattern with NF-κB expression. The expression of E-selectin mRNA and protein was positively correlated with the expression of NF-κB mRNA and protein. Conclusion Smoking can result in the high mRNA and protein expression of NF-κB and E-selectin in a dose dependent manner.
