甘氨酸对非酒精性脂肪肝小鼠瘦素水平的影响 被引量:2

摘要 目的甘氨酸对非酒精性脂肪肝小鼠瘦素(leptin)水平的影响。方法健康成年雄性昆明种小鼠40只,随机分4组:对照组、高脂模型组、甘氨酸(Gly)低剂量组、甘氨酸高剂量组,各组分别给予正常饲料和高脂饲料喂养2周,甘氨酸各剂量组隔日腹腔注射不同浓度Gly溶液。实验末,取肝组织观察小鼠非酒精性脂肪肝的形成,酶联接免疫法测定小鼠血清中leptin水平。结果光镜下高脂模型组肝组织可见弥漫性脂肪变性,甘氨酸各剂量组肝组织脂肪变性少,肝细胞内脂滴小且少,高脂模型组和甘氨酸各剂量组leptin水平明显大于对照组,甘氨酸各剂量组leptin水平显著高于高脂模型组,甘氨酸高、低剂量组之间leptin含量无显著性差异。结论 Gly对非酒精性脂肪肝形成有抑制作用,明显升高血中leptin水平可能是其作用机制之一。 Objective To investigate the effect of glycine on leptin level in nonalcoholic fatty liver mice. Methods Forty mice were randomly divided into four groups:control group,high fat diet group,low dose Gly group and high dose Gly group.The mice in the latter 3 groups were fed with high fat diet for 2 weeks.The mice in low dose Gly group and high dose Gly group were intraperitoneally injected with the glycine solution at a interval of a day.Leptin level in serum was detected by enzyme linked immunosorbent assay in the end of the experiment. Results The obvious diffuse hepatic steatosis was observed in high fat diet group,and slight hepatic steatosis in Gly groups.Serum leptin levels in high fat diet group and Gly groups were higher than that in control group(P〈0.01).Serum leptin level in Gly groups was higher than that in high fat diet group(P〈0.01),but there was no significant difference between low dose Gly group and high dose Gly group. Conclusion Glycine can prevent the nonalcoholic fatty liver by increasing leptin level in serum.
