组织芯片研究胃肠道间质瘤中E2F1、MDM2及P53的表达及意义

Expression and significance of E2F1,MDM2 and P53 in gastrointestinal stromal tumors:An immunohistochemical study using tissue microarray techniques

目的探讨E2F1、MDM2及P53在胃肠道间质瘤(gastrointestinal stromal tumors,GIST)中的表达及与GIST临床病理特征和预后的关系。方法利用组织芯片技术及免疫组化EnVision法检测123例GIST中E2F1、MDM2及P53的表达情况,分析三者表达与临床病理特征的关系,并对三者与GIST预后的关系进行统计学分析。结果 123例GIST中108例有三种蛋白均可评估,108例中E2F1、MDM2及P53蛋白三者阳性表达率分别为62.0%,43.5%和57.4%。三者表达均与NIH分级呈显著正相关(均P<0.001);在不同发生部位、是否坏死及有无转移复发之间差异均有显著性意义(均P<0.05),且三者表达在肠道发生的GIST明显高于胃和肠道外部位(P<0.05),胃和肠道外相比差异无统计学意义(P>0.05);而在不同发生年龄及不同细胞类型间相比差异均无统计学意义(均P>0.05)。结论 E2F1、MDM2及P53表达与GIST预后有关,三者对评价GIST预后是非常有意义的标记物。

Objective To investigate the expression of E2F1,MDM2 and P53 in gastrointestinal stromal tumors（GIST）and its relationship with clinical characteristics and prognosis. Methods EnVision immunohistochemical method and tissue microarray technology were used to detect E2F1,MDM2 and P53 expression in 123 cases of GIST.Their relationship with clinical characteristics and prognosis of GIST was analyzed. Results The positive expression rates of E2F1,MDM2 and P53 were 62.0%,43.5% and 57.4% in 108 cases of GIST,respectively. The positive expression of them was positively related to NIH grading of GIST（P0.001）,and was also related with tumor site,necrosis,metastasis and recurrence（P0.05）.The positive expression of them with the tumor in intestinal tract was higher than that in other sites（P0.05）.There was no significant difference in different age and cell category（P0.05）. Conclusion The expression of E2F1,MDM2 and P53 is closely related with the prognosis of GIST,and they may be significant molecular markers for prognosis of GIST.
