RP-HPLC法测定藁本内酯脂质体的体外释放度

Determination of drug release in vitro of ligustilide liposome by RP-HPLC

目的建立以反相高效液相色谱法测定藁本内酯脂质体的体外释放度的方法。方法采用溶出度第三法考察脂质体的体外释放过程。色谱条件为:色谱柱:ZORBAX-C18柱(4.6mm×250mm,5μm);流动相:甲醇-乙腈-水(34:20:46);流速1.0ml.min-1;检测波长274nm。结果药物的体外释放以对数正态分布方程拟合最好。结论该方法简便、易操作,可作为藁本内酯脂质体体外释药的研究。

Aim To establish a RP-HPLC method for the determination of drug release in vitro of ligustilide liposomes.Method The release behavior of the drug from liposomes was studied by the third method for dissolution.A ZORBAX C18 column（4.6 mm×250 mm,5 μm）was used with a mobile phase of methanol-acetonitrile-water（34：20：46）.The flow rate was 1.0 ml·min^-1,and the UV detection wavelength was set at 274 nm.Results Ligustilide release from liposomes in vitro fitted the log-normal distribution equation and had a property of sustained release.Conclusions The method is simple,fast and selective.It is suitable for determination of release profile in vitro of ligustilide liposomes.
