摘要
采用超高效液相色谱测定食品中的咖啡因,采用ACQUITYUPLC BEHC18色谱柱分离,以甲醇-水溶液为流动相进行洗脱,流速为0.4mL/min,柱温30℃,检测波长为272nm。结果表明,咖啡因与其他杂质均能达到基线分离,咖啡因的浓度在2~200mg/L时与峰面积成良好的线性关系。回收率为95.0%~99.1%,精密度测试结果的RSD为1.02%。该方法简便、准确、快速、重现性好、回收率高,适用于食品中咖啡因的测定。
Ultra performance liquid chromatography(UPLC)was used for the simultaneous determination of caffeine in food.Chromatographic separation was performed on ACQUITY UPLC BEH C18 column.The mobile phase was a mixture of methanol and deionized water at a flow rate of 0.4 mL/min and the column temperature was 30℃.The UV detection wavelength was fixed at 272 nm.Results showed that caffeine and other compounds were separated well and the standard curve of caffeine had good linearity in the concentration range of 2~200 mg/L.The recovery rate was 95.0% ~ 99.1%.The RSD was 1.02%.This method was proved to be simple,accurate,rapid with good reproducibility and high recovery rate.It was suitable for the determination of caffeine in food.
