不同环境对局灶性脑梗死大鼠梗死灶周围皮层神经丝蛋白表达的影响 被引量:1

Effects of Different Environmental Intervention on Neurofilament Expression in Rats after Unilateral Local Cerebral Infarction

摘要 目的观察不同环境对局灶性脑梗死大鼠梗死灶周围皮层神经丝蛋白(NF)表达的影响。方法 SD大鼠125只,电凝法造成右侧大脑中动脉阻断(MCAO)模型后随机分为独居组(n=30,独居于标准笼),社交组(n=30,5只一组群居于标准笼),探索学习组(n=30,15只一组居于迷宫笼),丰富环境组(n=30,5只一组居于丰富环境笼)和假手术对照组(n=5,5只一组居于标准笼)。各试验组分别于1d、3d、1周、2周、3周、4周各组随机选取5只大鼠处死,测定皮层梗死灶周围区NF光密度(OD)。结果探索学习组及丰富环境组在1周后各时间点NF光密度均明显高于其他两组(P<0.01)。结论丰富环境及探索学习均能促进局灶性脑梗死大鼠梗死灶周围皮层NF的表达。 Objective To observe the effects of different environmental intervention on neurofilament (NF) expression in rats after unilateral local cerebral infarction.Methods After middle cerebral artery occluded (MCAO) by electric coagulation,125 male SD rats were randomly divided into individual living group (n=30,living alone in small standard cages),social communication group (n=30,5 as a group living in large standard stages ),learning group(n=30,15 as a group living in exploratory cages),enriched environment group (n=30,5 as a group living in EE cages) and sham operated group(n=5).The rats were randomly sacrificed at the 1st,3rd,7th,14th,21st,28th day after MCAO.The expressions of NF in peri-ischemic cortex were detected with immunohistochemistry staining.Results The expression of NF in the peri-ischemia cortex in enriched environment group and learning group was higher than that in other two groups (P0.01) after 7 days,it also was higher in social communication group than that in individual living group (P0.05).Conclusion Enriched environment and learning could enhance NF expression in rats after unilateral local cerebral infarction.
