PDX-1蛋白对TNF-α和IL-1β诱导的大鼠胰岛凋亡的保护作用 被引量:1

摘要 目的探讨PDX-1蛋白对细胞因子(TNF-α、IL-1β)诱导的大鼠胰岛凋亡的保护作用。方法 PDX-1基因克隆及表达质粒的构建,PDX-1蛋白的表达、纯化和鉴定,PDX-1蛋白保护TNF-α和IL-1β引起的胰岛细胞凋亡的研究。结果 PDX-1蛋白可以延缓胰岛细胞的自然凋亡,降低处理后的胰岛Caspase-3活性和荧光细胞的数目;PDX-1蛋白干预后的细胞因子组的葡萄糖刺激胰岛素释放为(1.586±0.129)μI U/ml,与对照组(1.375±0.131)μI U/ml,相比有显著差异(P<0.05)。结论表明PDX-1蛋白可以减少细胞因子诱导的胰岛凋亡,保护β细胞功能。 Objective To study whether the PDX-1 protein has antiapoptosis effects induced hy cytokine on rat islet of Langerhans. Methods PDX-1 gene cloning and expression plasmid construction, PDX-1 protein expression, purification and identification, study of PDX-1 proteins counteracts apoptosis on primary rat islet of Langerhans induced by TNF-α and IL-1β were used. Results PDX-1 protein could delay the natural apoptosis of rat islet, and decrease Caspase-3 activity and the fluorescent cells. PDX-1 protein could promote an increase in insulin secretion (1. 586 ±0. 129) μIU/ml compared with control group of(1. 375±0. 131) μIU/ml (P〈0. 05). Conclusion PDX-1 proteinscould counteract apoptosis on primary rat islet of Langerhans induced by TNF-α andlL-1β.
