摘要
目的:探讨建立静注内毒素致麻醉状态下绵羊急性呼吸窘迫综合征(ARDS)模型的方法。方法:绵羊静脉给予内毒素(LPS)3μg/kg,30分钟匀速泵入。观察血流动力学、肺力学参数和肺气体交换的变化。结果:12只绵羊中有8只在3小时内发生ARDS。第4~8小时间血流动力学(心脏指数、平均血管阻力指数、肺血管阻力指数、氧输送)、肺力学参数(肺动态顺应性、平均气道阻力、气道峰压值、气道阻力)、肺气体交换(改良氧合指数、动脉氧分压)均平稳(P均>0.05)。结论:静注LPS能够复制出稳定时间较长的麻醉状态下绵羊ARDS模型。
Objective:To establish an acute respiratory distress syndrome (ARDS)model in sheep under anesthesia state by intravenous lipopolysaccharide(LPS) challenge.Methods:Sheep was infused intravenously with LPS (3 μg/kg) within 30 minutes,and parameters of hemodynamics,lung mechanics and gas exchange were observed during the experiment.Results:8 of 12 sheep developed typical ARDS within 3 hours following LPS challenge.All parameters were kept constantly from the fourth to the eighth hour (all P >0 05), inclu ding hemodynamics (CI,O 2,PVRI,SVRI),lung mechanics (dynamic compliance,resistance,peak inspiratory pressure,mean airway pressure) and gas exchange (PaO 2/FiO 2,PaCO 2).Conclusions:A prolonged ARDS sheep model could be set up by intravenous LPS challenge under anesthesia state.
