摘要
目的:观察复方葶苈注射液对体外新生小牛肺主动脉内皮细胞单层通透性的影响。方法:新生小牛肺主动脉内皮细胞培养于醋酸纤维膜上,分别测定Hank′s液1∶10稀释的复方葶苈注射液作用60分钟前后同一滤膜上正常内皮单层和组胺处理内皮单层的滤过系数(Kf)和渗透压反射系数(σ)。结果:经1∶10的复方葶苈注射液作用后,正常内皮单层〔(11.80±0.75)μl·min-1·cm-2·kPa-1比(7.77±0.67)μl·min-1·cm-2·kPa-1〕和组胺处理内皮单层〔(16.87±1.11)μl·min-1·cm-2·kPa-1比(12.82±1.19)μl·min-1·cm-2·kPa-1〕的Kf均明显降低(P均<0.01),σ(正常内皮单层σ为0.32±0.06比0.44±0.03,组胺处理内皮单层σ为0.19±0.06比0.28±0.06)则均明显升高(P均<0.01)。结论:复方葶苈注射液有降低肺主动脉内皮单层通透性的作用,可减轻肺动脉高压引起的肺水肿。
Objective:To observe the influence of Tingli compound injection on the permeability of in vitro cultured endothelial cell monolayer.Methods:The endothelial cells prepared from newborn calf pulmoaorta were cultured on cellacefate filters to form cell monolayer.The filtration coefficients ( Kf ) and the osmotic reflection coefficients ( σ ) of the cell monolayer on the same filter,treated with or without histamine,were determined before and after 60 minutes incubation with 1∶10 Tingli compound injection.Results:The filtration coefficients of the endothelial cell monolayer,both with or without treatment with histamine,were significantly decreased (both P <0 01) and the osmotic reflection coefficients were markedly increased (both P <0 01).Conclusions:Tingli compound injection has the beneficial effect on reducing permeability of endothelial cell monolayer and may attenuate lung edema caused by pulmonary hypertension.
