UPLC测定山茱萸生品与酒炙品中齐墩果酸与熊果酸的含量

Determination of Oleanolic Acid and Ursolic Acid in Crude and Processed Fructus Corni by UPLC

目的:建立山茱萸药材中齐墩果酸和熊果酸的超高效液相色谱含量测定方法。方法:采用超高效液相色谱法,ACQUITYUPLCTMHSSC18色谱柱(2.1mm×100mm,1.8μm),流动相:甲醇-0.1%甲酸水(85∶15),流速:0.2mL·min-1,检测波长:210nm。结果:齐墩果酸和熊果酸分别在0.010～0.500mg.mL-1(r=0.9999)、0.010～0.500mg.mL-1(r=0.9999)与峰面积呈良好的线性关系,平均回收率分别为100.4%、94.4%(n=9),RSD均为1.8%。结论:该方法快速、简便、准确,可用于山茱萸药材的质量控制。

Objective ： To develop a method for the determination of the contents of oleanolic acid and ursolic acid in crude and processed Fructus Corni by UPLC. Methods： An ACQUITY UPLC^TM HSS C18 column （2. 1mm × 100mm,1.8μm） was used with methanol-0. 1% formic acid water （85：15） as the mobile phase . The flow rate was 0. 2mL·min^-1 and the detection wavelength was 210nm. Results： The calibration curve showed good linearity relationships in the ranges of 0. 010 -0. 500 mg·mL^-1 （r = 0. 999 9） and 0. 010 -0. 500mg·mL^-1 （r =0. 999 9） for oleanolic acid and ursolic acid, respectively. Average recoveries of oleanolic acid and ursolic acid were 100. 4% with RSD = 1.8% （n = 9） and 94. 4% with RSD = 1.8% （ n = 9 ）, respectively. Conclusion： This method was simple, fast, accurate and reproducible, which was suitable for the quality control of Fructus Corni.
