摘要
目的:研究大鼠骨髓干细胞移植后的定位与分化状态。方法:以荧光染料DiI为示踪剂对移植的骨髓干细胞进行标记,并通过门静脉移植入大鼠肝脏,分别于7、14、21、28d采集移植鼠的肝组织进行荧光检测。结果:荧光剂DiI标记的干细胞在移植鼠肝组织中定位,并且分化增殖。其亮红色荧光随时间的推移略有减弱,但并不消失。结论:DiI荧光剂为可靠的干细胞示踪剂,可应用于移植细胞的标记,骨髓干细胞可在受损肝组织中定位并分化增殖。
Objective:To research the location and differentiation of rats bone marrow stem cells after transplantation.Methods:Labeled rats bone marrow stem cells with fluorochrome DiI, transplanted the labeled cells into the rat livers via portal vein, collected and detected the hepatic tissues from the transplantation rats after 7,14,21,28 days respectively.Re-sults:The stem cells labeled by fluorochrome DiI located, proliferated and differentiated in the livers of the transplantation rats.The red fluorescence declined over time, but it didn't disappear.Conclusion:DiI is a reliable stem cell tracer and can be used to label transplanted cells.Bone marrow cells can be located proliferated and differentiated in damaged hepatic tissue.
