牛磺胆酸对小鼠腹腔巨噬细胞功能的影响

摘要 为了阐明TCA对小鼠巨噬细胞吞噬功能的影响,本研究分别采用鸡红细胞半体内法和体外培养的腹腔巨噬细胞吞噬中性红法测定了TCA对体内、外小鼠腹腔巨噬细胞吞噬功能的影响,结果证明,高、低剂量的TCA均能极显著的提高小鼠腹腔巨噬细胞吞噬鸡红细胞百分率(P<0.01),显著提高小鼠腹腔巨噬细胞的鸡红细胞吞噬指数;0.015μg/ml、0.3μg/ml、1.5μg/ml和15μg/ml的TCA均能显著提高体外培养小鼠腹腔巨噬细胞对中性红的吞噬率(P<0.05),表明TCA具有提高巨噬细胞吞噬功能的作用。采用ELISA法测定了TCA对小鼠巨噬细胞分泌白介素-1β的影响,试验结果表明,高、低剂量的TCA均能极显著提高正常和免疫抑制小鼠血清中的含量(P<0.01),1.5和15?g/mL的TCA可显著提高体外培养小鼠腹腔巨噬细胞IL-1β的分泌量(P<0.01),0.015μg/ml和1.5μg/ml的TCA可极显著提高LPS活化的小鼠腹腔巨噬细胞IL-1β的分泌量(P<0.01),0.3μg/ml和15μg/ml的TCA可显著提高LPS活化的小鼠腹腔巨噬细胞IL-1β的分泌量(P<0.05),表明TCA具有促进腹腔巨噬细胞的分泌IL-1β的功能。 For clarifying the effects of TCA on phagocytosis of macrophage from mice,the effects of taurocholic acid(TCA) on phagocytosis of macrophage from mice in vivo and vitro was studied using the CRBC semi-intracorporal method and toluylene red phagocytose method.The results showed that TCA could significantly increased the rate of the peritoneal macrophage phagocytizing CRBC(P0.01).TCA also could significantly increased cytophagic index(P0.05).In addition,the phagotrophic rate of peritoneal macrophage cultured in vitro phagocytizing toluylene red was significantly increased when exposed with TCA at 0.015,0.3,1.5 and 15?g/mL,respectively(P0.05).The results suggest that TCA increase the phagocytosis of macrophage.The effects of TCA on mice macrophage for secretion of IL-1β was determined by ELISA,the results showed that TCA at low and high dose both could increase the IL-1β level in blood serum of normal and immune suppressed mice(P0.01).The level of plasma IL-1β from mice peritoneal macrophage cultured in vitro was significantly increased cultured with1.5 and 15?g/mL TCA(P0.01).The level of plasma IL-1β from mice peritoneal macrophage which activated with LPS could significantly increased by 0.015,1.5,0.3 and 15?g/mL TCA,respectively.The present results indicate that TCA could stimulate the secretion of IL-1β in mice macrophage.
