摘要
目的研究外用绞股蓝紫对紫外线照射引起的无毛小鼠皮肤组织线粒体DNA损伤的作用。材料与方法将昆明种无毛小鼠随机分成5组:正常对照组、照射组、照射加绞股蓝低、中、高剂量组。模拟日光中UV长期照射,给药组照射同时外用绞股蓝。测定皮肤组织中线粒体DNA缺失情况。结果所有模型组小鼠皮肤组织中均存在mtDNA(3867bp)大片段缺失,而正常对照组以及给药组中的中、高剂量组小鼠皮肤组织中mtDNA(3867bp)大片段缺失的发生率和缺失占总mtDNA的百分比均显著低于照射组。结论绞股蓝可防止由紫外线照射引起的线粒体DNA损伤,对紫外线照射下的无毛小鼠皮肤具有保护作用。
Objection To research the effect of the external used Gynostemma Pentaphyllum to the damaged cytochondriome DNA in the skin tissue of the uviolized hairless mouse. Material & Method The Kunming hairless mice were randomly divided into 5 groups: normal control, irradiated group, 3 MAT groups (LD, MD, HD). All the 5 groups were long-term irradiated by stimulating the UV in the sunshine, and meanwhile the mice of the 3 MAT groups were treated by the external used Gynostemma Pentaphyllum. Finally, we determined the depletion of cytochondriome DNA in the skin tissue. Result The large fragment deletion of mtDNA(3867bp) exist in all the 5 groups. The incidence rate and the percentage of the mtDNA (3867bp) deletion in the Normal control and the 3 MAT Groups is obviously lower than that in irradiated group. Conclusion Gynostemma Pentaphyllum can prevent the cytochondriome DNA in the skin tissue from the damage of the ultraviolet irradiation and protect the skin of the uviolized hairless mouse.
