摘要 以杨树叶片为外植体,MS为基本培养基,使用不同激素不同浓度配比,建立了杨树叶片离体培养植株再生体系。结果表明:叶片的最佳诱导培养基为:MS+2,4-D 1.0 mg/L+6-BA 1.0 mg/L;不定芽培养基为:MS+NAA1.0 mg/L+6-BA0.5 mg/L。NAA与BA对愈伤组织诱导有调控作用,NAA/BA比值增大有利于不定芽的诱导,但不能超过4∶1;BA与2,4-D关系比较复杂,当2,4-D和6-BA的浓度均为1.0mg/L时,愈伤组织诱导效果最好。 The tender leaves of Populus euramericana 108 as the explants were cultured in the MS basic medium supplemented with different concentrations of plant growth regulators to establish the in vitro regeneration system.The results showed that the optimal induction medium was MS+2,4-D 1.0 mg/L+6-BA 1.0 mg/L and the medium suitable to induce the adventitious buds was MS+NAA 1.0 mg/L+6-BA 0.5 mg/L.NAA and BA had the regulation effects on the induction of calli,that was to say,the increasing ratio not more than 4∶1 was beneficial to induce the adventitious buds;while the relationships of BA with 2,4-D were more complex,the induction effect was the best with 2,4-D and 6-BA of 1.0 mg / L,respectively.
