摘要
目的:研究罗勒提取物抗血栓作用并从抗凝、纤溶两个方面探讨其作用机制。方法:采用小鼠肺血栓模型、FeCl3诱导大鼠颈动脉血栓模型及大鼠下腔静脉血栓模型3种实验性血栓模型,考察罗勒水提物与醇提物的抗血栓形成作用;并通过采用酶联免疫吸附测试法(ELISA)测定动物血浆AT-Ⅲ、PC及PLG、t-PA、PAI-1几个指标观察各组药物对抗凝及纤溶活性的影响进而探讨其作用机制。结果:罗勒提取物可显著提高胶原蛋白-肾上腺素混合诱导剂所致肺血栓小鼠的存活率,明显延长FeCl3诱导大鼠颈动脉血栓形成时间并显著减轻血栓湿重,显著减轻结扎法所致下腔静脉血栓湿重,并使血浆AT-Ⅲ浓度降低、PC活性升高,PLG、t-PA活性升高、PAI-1活性轻微降低、t-PA/PAI-1比值升高。结论:罗勒提取物无论是水提物还是醇提物均有明显抗血栓作用,其作用机制可能与抗凝系统和纤溶系统活性升高有关。
AIM:To study the antithrombotic activity of Ocimum basilicum L(OBL) extracts,extracted with aqueous and 70% ethanol,and to observe anticoagulat action and fibrinolysis. METHODS:Three experimetal models in vivo,pulmonary thromboembolism in mice,FeCl3-induced arterial thrombosis and ligation-induced inferior caval thrombosis in rats were used to examine the antithrombotic activity of aqueous and alcohol extract of OBL ; the activity of AT-Ⅲ,PC,PLG,t-PA and PAI-1 in plasma were determined by ELISA to evaluate the effects of thedrugs on the anticoagulant factor and fibrinolytic system. RESULTS: OBL extract by oral administration could markedly increase the survival rate of the mouse suffering from pulmonary thrombosis induced by the mixture of collagen and epinephrine,and significantly decreased the weight of the thrombus induced by ligation of inferior vena cava. The concentration of AT-Ⅲ was decreased,the concentration of PC was increased; the activity of PLG and t-PA were increased,the activity of PAI-1 was decreased slightly,the ratio of t-PA/PAI-1 increased. CONCLU-SION: The OBL extracts,both aqueous and alcholol extract,have strong anti-thrombosis effect,the action of mechanism may have correlation with the increase in the activity of the anticoagulant factor and fibrinolysis system.
