摘要 目的:观察黑木耳多糖(Auricularia auricula polysaccharide,AAP)体外对人红白血病K562细胞的抗肿瘤作用及其机制。方法:将K562细胞与AAP以及AAP作用于体外培养人外周血单个核细胞(PBMCs)的上清液培养,用MTT法检测K562细胞增殖和流式细胞仪检测细胞周期和凋亡情况。结果:AAP对K562细胞没有显示直接的细胞毒作用,但AAP作用的PBMC上清液能显著抑制K562细胞的增殖,抑制率最大可达49.68%;AAP作用的PBMCs上清液能使G0/G1期细胞显著增加(P<0.05,P<0.01);200μg/mL到800μg/mL浓度AAP刺激的PBMCs上清液能显著诱导K562细胞凋亡(P<0.05,P<0.01),其凋亡率可达24.70%。结论:AAP通过激活免疫细胞抑制肿瘤细胞生长,诱导其凋亡并使细胞阻滞于G0/G1期。 AIM: To identify the antitumor effect of Auricularia auricula polysaccharide(AAP)on human erythroleukemia cell line (K562) and its mechanism in vitro. METHODS:K562 cells were cultured with AAP alone and in common with the supernatant of peripheral blood mononuclear cells (PBMCs) together with AAP,and the proliferation of K562 cells was determined with MTT assay. The flow cytometry was applied to investigating the cell cycle and apoptosis of K562 cells. RESULTS: AAP itself did not show any direct cytotoxicity to K562 cells. The supernatant of PBMCs treated with AAP revealed inhibitory effect on the cell proliferation up to 49. 68% growth inhibition rate. The K562 cells at G0 /G1 phase were markedly extended(P 0. 05,P 0. 01),whereas S,G2 /M phases were significantly shortened. The apoptosis rate of K562 cells was significantly increased at the concentration of AAP at 200 μg/mL to 800 μg/mL(P 0. 05,P 0. 01),and the apoptosis rate reached as high as 24. 70% . CONCLUSION: AAP can activiate immune cells to inhibit the growth of tumour,induce tumour cell apoptosis and make the cell cycle stop in G0 / G1 phase.
