荧光定量PCR检测山羊褪黑素受体表达体系的优化研究 被引量:1

摘要 本研究通过系统对比试验,优化建立了检测山羊卵巢组织中褪黑素受体基因表达量差异的荧光定量PCR技术体系,在这种体系下检测褪黑素受体1A基因在山羊卵巢组织中的差异表达量,可以得到最优的标准曲线,使起始模板浓度与循环数有良好的线性关系,且扩增效率百分数Eff接近1;荧光动力学曲线符合标准的"S"形荧光增长曲线,曲线拐点清楚,基线平而无明显上扬趋势;熔解曲线成单峰,充分说明扩增的特异性高,产物单一,与SYBR Green I荧光染料结合的为目标DNA片段,荧光曲线能够准确反映目的产物的扩增结果。
