摘要 目的:探究1-磷酸鞘氨醇(S1P)预处理能否激活Akt1而保护缺氧复氧心肌细胞。方法:原代培养大鼠乳鼠心肌细胞,制备缺氧/复氧心肌细胞损伤模型,应用台盼兰排斥法和Western blot分别检测S1P预处理对心肌细胞死亡率和磷酸化Akt1(p-Akt1)水平的影响。结果:S1P预处理能够明显降低缺氧/复氧心肌细胞死亡率,提高缺氧/复氧心肌细胞磷酸化Akt1水平。结论:S1P预处理可能通过激活Akt1而保护缺氧复氧心肌细胞。 Objective: To explore whether pretreatment of SIP can activate Aktl to protect hypoxia-reoxygenationin cardiomyocytes. Methods, Culture neonatal rat cardiomyocytes, build the model of hypoxia-reoxygennation, the impact of pretreatment of SIP on cell mortality rate and p-Akt were studied by using trypan blue staining and western blot. Results: Pretreatment of SIP can reduce cell mortality rate and improve the level of p-Akt1 in hypoxia-reoxygenation cardiomyocytes significantly. Conclusion. Pretreatment of SIP can activate Akt1 to protect hypoxia-reoxygenationin cardiomyocytes.
