波动性高糖对人脐静脉内皮细胞ICAM-1及TNF-α表达的影响 被引量:1

摘要 目的探讨波动性与持续性高糖对人脐静脉内皮细胞(HUVECS)细胞间黏附分子-1(ICAM-1)和肿瘤坏死因子(TNF-α)表达的影响。方法以HUVECS为研究对象,实验分为3组:正常对照组(5.5mmol/L)、持续高糖组(25mmol/L)、波动高糖组(5.5/25mmol/L,白天3h高糖2h正常浓度葡萄糖波动3次,高糖过夜)。通过双抗体夹心酶联免疫吸附法(ABC-ELISA)检测细胞在各种糖浓度条件下24h、48h、72hICAM-1及TNF-α的表达。结果ICAM-1的浓度各个时间段波动组与高糖组和正常对照组相比均升高(P<0.05),呈时间-效应关系。TNF-α的浓度3个时间段波动组与高糖组、正常对照组相比显著升高(P<0.05),呈时间-效应关系。结论波动性高糖较持续性高糖更增强HUVECSICAM-1和TNF-α的表达。
